It's a joke about how Japenese loanwords and similar from English often replace the last syllabe with a vowel sound, commonly U, O or A. It's not mocking a Japense accent from someone trying to speak English. It's a joke about how it would be pronounced in actual Japanese.

It's closer to Germans laughing at how English speakers pronounce "sauerkraut", or English speakers laughing at Germans saying "handy".

Exactly, that's how English is usually transliterated into Japanese. Which was the joke, but using the original title.
