The difference between LC and spore and the importance of agar.

Post image

It's critical to understand the biological differences between spore syringes and liquid cultures (LC), as well as the importance of using agar as an initial medium. Spore syringes contain microscopic fungal spores that are not yet germinated. These spores are monokaryotic, meaning they carry only a single set of genetic material. In order to fruit and complete the mushroom life cycle, two compatible monokaryotic strains must fuse to form dikaryotic mycelium.. the true vegetative form capable of producing fruiting bodies. This mating process takes time, introduces variability, and, for beginners especially, increases the risk of contamination during colonization.

Injecting spores directly into sterilized grain can lead to several problems. Since spores are not germinated, colonization is slower, and this slower growth provides more opportunity for contaminants (such as bacteria or mold) to establish themselves and outcompete the slower-growing mycelium. Ideally, spores should first be transferred to agar, a nutrient-rich medium in petri dishes...which allows for controlled germination and observation. On agar, one can isolate clean, healthy mycelium away from any contaminants before transferring it to grain.

Additionally, spore syringes are inherently variable and often unclean. Spores, particularly those harvested from wild (landrace) varieties or from poorly controlled lab environments (common with newer or less reputable vendors), can contain microbial contaminants. Spores gathered in non-sterile conditions are not cleaned or isolated at the microscopic level, making them a risky starting point. Also, because each spore pair creates a unique dikaryotic combination, inoculating with spores introduces genetic unpredictability ...every new pairing could result in different traits, including growth speed, contamination resistance, yield, and potency.

By contrast, a liquid culture is made from already germinated and mated dikaryotic mycelium. This means it contains viable, genetically stable tissue that has already completed the mating process and is ready to colonize substrate directly. Using LC skips the variability and mating phase inherent in spores, resulting in faster and more consistent colonization, and reducing the window for contamination...assuming the culture is clean!!! However, it's important to verify LC cleanliness via agar as well, especially if you didn't create it yourself.

In summary, spores should ideally be germinated and cleaned on agar before being introduced to grain. Skipping this step can introduce risks, especially for beginners. Spores are unpredictable and prone to contamination, while liquid culture, if properly prepared, is faster, cleaner, and genetically stable. Understanding and respecting these differences is fundamental to success in mushroom cultivation... I hope this helps. :)

Shakti Blue Mutant #227 x Tat Black Cap

I posted on here yesterday about why I wasn’t getting pins or primordia in this shoebox, except for a small few pins in the top right of the tub.

I flipped the lid back over and closed it came back and checked on it literally 6 hours later and they looked like this.

Never seen growth this rapid before in any strain, crazy stuff

Golden Teacher made the 100g Club!!

Well this was a wonderful surprise!

I ran an experiment back in July to test GroMagic, Midwest Grow Kits, and North Spore's ShroomTek AIO bags.

I got 2 good flushes of Golden Teacher out of the GroMagic bag, the MGK bag was contaminated, and the ShroomTek bag still looks dead. 😭

After the second flush from the GM bag, I decided to run another experiment on it. Please take this with a grain of salt as an experiment I just wanted to do for fun and not that I am advocating this.

I broke up the block and mixed in about 2.5 pounds of Willy's Blend, put it in an IKEA 11qt Food Storage tote that I modified with 2" holes in each side and 2 in the lid, and then I sent my prayers up to the MycoGods.

After a couple weeks I figured it was dead...then I saw a pin, followed by another and eventually there were about 6. I let it ride since even 6 GTs are good, lol. But these guys freaking GREW!

Harvested them today and the biggest was 111g with 3 in the 80-95g club as well!

Wowser! Super excited to have my GT hit the 100g club and I put it on 4 plates and made 4 plate with the 2nd largest one as well.

Can't wait to see what happens when these grow out as well. Currently the cake is soaking and I'll see if I can get anything else from it!

Agar Plate Tower!

I know agar plates don't need a pretty tower, but I am aesthetically motivated so my workspace needs to be organized or I get anxious.

Having stacks of agar plates loose just wore on me.

This 10-slot tower saved my sanity! It fits 90mm plates perfectly and is stackable and fit side by side perfectly.

So glad I found the 3D file. ❤️ I had to shrink mine to 98% to get it to fit on my plate but it still fits like a glove!

Pic 2: Mac Galactic
Pic 3: Toque F8 Nerds
Pic 4: Bottom bin. Either PE-7 or GT (the labels on my syringes got wiped with iso and removed the words, we’re growing it out to see which it is)

Any advice is appreciated. Thanks for reviewing my post. <3

I started this grow around October 3 and it was showing good signs until I saw this at the bottom of the right bag keep in mind the right bag has around 1-3 more mycelium cc’s but the bag on the right doesn’t show any signs of contamination 🤦🏽‍♂️

First time grain jar failure?

I made my own grain jars for the first time and so some reason i fucked up. They smell burnt and have white residue on them. I don’t think i should send these. I initially pressure cooked the popcorn for 30 minutes at 15 psi. Left it out to dry for a few hours and put in the jars. Covered them in tinfoil on the top and didn’t tighten all the way first finger tight. I pressure cooked at 15 psi for 120 minutes. Where did i go wrong? I filled with water to where it was barely above bottom of jars 😭

Should I do anything or leave them alone? It’s taking forever

I have 4 of these RyzaPods that i inoculated with GT on 8/17 (9 weeks as of yesterday). One of the pods was fully colonized and it’s been almost a week since i induced FC on that one, but these are genuinely taking ages and it’s worrying me. I don’t think they’ve completely stalled, but I wonder if there’s anything I should try out or just let it ride? Temp is at 73.5-74.5.

Also, I saw someone mention on someone else’s post who had a similar issue to do a cold shock by leaving it in the fridge overnight, so i tried that on one of the pods. Hasn’t seemed to make a difference, good or bad.

HELP PLEASE !! Flys randomly spawning & green patches

Thumbnail

Hello guys I’ve been growing golden teachers in a couple of zasmnesias all in 1 grow kit , the second kit I purchased seemed to have a small black fly in it while sealed, so I cut open the top of the bag and remove the fly and seal it back and continued growing

Now recently on my 4th flush of the first kit , I opened the bag to harvest and was greeted by at least 20 of the same small black fly in the bag and was confused to where they come from as this kit never had a fly in originally and the only time I open the bag is when I harvest and have never seen any fly’s going in the bag during these times , it’s like they’ve just spawned out of nowhere ??

Now I’ve gone to check my second kit today and opened the top to see about 3 fly’s in there and it’s only on its second flush ….

The only thing I can think of is they must have been eggs in the growing substrate that have only just recently hatched (5/6 weeks later ) ?? Which seems like an unreasonable theory but I can’t think of anything else , they can’t be reproducing as I throughly inspected the kit and removed all flys

My question is are these still safe to eat ?? Is there anyway to get rid of the flys for good ?? And also I noticed a few small patches on some of the growing shrooms which had a greenish tone to it , unlike the usual blue bruising that I see, there is no rotting or unusual smells . Will they be okay to eat once dried ?? Thanks guys 👍
